Consensus represents the second generation of protein structure prediction tools. It uses an automated method for analyzing the structures of a family of homologous proteins, proposing a series of possible conformations consistent with the available data. BIOSYM's Homology and Consensus programs form a synergistic duo to achieve the goal of predicting a reliable model structure from sequence data.
The Consensus program examines and analyzes conformations of reference proteins. It generates constraints consistent with the geometries of their conserved regions, along with standard, covalent constraints for the variable regions. Unlike traditional homology modeling methods, structural information from all of the reference proteins is used at once, and with equal weight. No single reference protein's conformation is used alone.
